Multicolor flow cytometric analysis of CD38 expression on BALB/c mouse spleen. Splenocytes were stained simultaneously with PE Hamster anti-Mouse CD3 antibody (Cat. No. 553063/553064/561824) and with either Alexa Fluor® 647 Rat IgG2a, κ Isotype Control (Cat. No. 557690; Left Panel) or Alexa Fluor® 647 Rat anti-Mouse CD38 antibody (Cat. No. 562769; Right Panel). Two-color flow cytometric dot plots show the correlated expression patterns of CD38 (or Ig isotype control staining) versus CD3 for gated events with the forward and side light-scatter characteristics of viable spleen cells. Flow cytometry was performed using a BD™ LSR II Flow Cytometer System.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
  4. The Alexa Fluor®, Pacific Blue™, and Cascade Blue® dye antibody conjugates in this product are sold under license from Molecular Probes, Inc. for research use only, excluding use in combination with microarrays, or as analyte specific reagents. 90/CD38

The 90 monoclonal antibody specifically binds to CD38, a 42 kDa transmembrane glycoprotein on immature and mature, resting and activated, B lymphocytes. In contrast to humans, CD38 expression is down-regulated on mouse germinal center B cells and plasma cells. CD38 is also expressed on a subpopulation of thymic and peripheral T cells, NK cells, and splenic macrophages. Furthermore, CD38 has been detected on bone marrow-derived hematopoietic stem cells. The CD38 molecule is reported to exhibit both cyclase and hydrolase activities and plays a role in lymphocyte activation. CD31, both human and mouse, is reported to be a ligand for human CD38.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
