Flow cytometric analysis of CD27 expression on Rhesus macaque (Macaca mulatta) peripheral blood lymphocytes. Rhesus whole blood was stained with either FITC Mouse Anti-Human CD27 (Cat. No. 557329/555440/560986; solid line histogram) or FITC Mouse IgG1, κ Isotype Control (Cat. No. 555649; dashed line histogram). Erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). Fluorescent histograms were derived from gated events with the side and forward light-scattering characteristics of viable lymphocytes.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with FITC under optimum conditions, and unreacted FITC was removed.

Product Notices

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Source of all serum proteins is from USDA inspected abattoirs located in the United States.
  4. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

Clone M-T271 reacts with the human form of 110 kDa disulfide-linked dimer of two polypeptide chains. CD27 molecule is a lymphocyte-specific member of the TNFR/NGFR family, and is expressed on the subset of human thymocytes and on the majority of mature T lymphocytes. This clone also cross-reacts with a major subset of peripheral blood lymphocytes and monocytes of baboon, and both rhesus and cynomolgus macaque monkeys. CD27+ cells were detected on all CD3+ lymphocytes and on a subset of CD20+ cells. The reactivity on lymphocytes and monocytes is similar to that observed with peripheral blood lymphocytes from normal human donors.

Fluorescein (FITC) is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 518-nm. FITC is designed to be excited by the Blue laser (488-nm) and detected using an optical filter centered near 520 nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
