Alexa Fluor® 647 Mouse anti-Human CD192 (CCR2)
Flow cytometric analysis of Alexa Fluor® 647-conjugated anti-human CCR2 on human peripheral monocytes. Human peripheral leukocytes were stained with Alexa Fluor® 647-conjugated anti-human CCR2 antibody (Clone 48607, Cat. No. 557913; open histogram) or Alexa Fluor® 647-conjugated mouse IgG2b (Clone 27-35, Cat. No. 557903; shaded histogram).  Flow cytometry was performed and the histograms were derived from the gated events based on light scattering characteristics of viable monocytes.

Preparation and Storage

The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Product Notices

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

  4. Alexa Fluor® 647 fluorochrome emission is collected at the same instrument settings as for allophycocyanin (APC).

  7. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The 48607 monoclonal antibody specifically binds to CD192 which is also known as, Chemokine C-C motif receptor 2 (CCR2), Monocyte chemoattractant protein 1 receptor (MCP-1 receptor/MCP-1-R), or CKR2. CD192 is a seven-transmembrane, G-protein-coupled receptor that is the specific receptor for Monocyte chemoattractant protein 1 (MCP-1/CCL2), MCP-2/CCL8, MCP-3/CCL7, and MCP-4/CCL13. CD192 is expressed on monocytes, dendritic cells, NK cells, basophils, and activated T cells and B cells. CD192 exists in two forms, CD192A and CD192B. The two forms are derived from alternatively spliced variants of a single gene and differ at the C-terminal end. CD192 plays an important role in inflammatory responses.

Alexa Fluor™ 647
Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
