Western blot analysis of Caspase-11 protein expression by mouse J774 cells. Lysate was prepared from cells of the mouse J774 macrophage cell line. Cell lysate proteins were resolved under reducing conditions by electrophoresis (SDS-PAGE) in a 4-20% Tris-Glycine polyacrylamide gel and transferred to a PVDF membrane. The blot was then probed with either 0.250 µg/ml (Lane 1), 0.125 µg/ml (Lane 2), or 0.063 µg/ml (Lane 3) of Purified Rat Anti-Mouse Caspase-11 antibody (Cat. No. 564971) followed by detection with HRP-conjugated Goat Anti-Rat Ig (Cat. No. 554017), and ECL Western blot detection reagents. Caspase-11 was identified as protein bands of approximately 38 to 43 kDa.

- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
- Sodium azide is a reversible inhibitor of oxidative metabolism; therefore, antibody preparations containing this preservative agent must not be used in cell cultures nor injected into animals. Sodium azide may be removed by washing stained cells or plate-bound antibody or dialyzing soluble antibody in sodium azide-free buffer. Since endotoxin may also affect the results of functional studies, we recommend the NA/LE (No Azide/Low Endotoxin) antibody format, if available, for in vitro and in vivo use.

The 17D9 monoclonal antibody specifically binds to mouse Caspase-11 (CASP-11/Casp11) which is also known as ICH-3. Mouse Caspase-11 is homologous to human Caspase-4 and is encoded by the mouse Casp4 (Caspase 4, apoptosis-related cysteine peptidase) gene. Caspase-11 is a cysteine protease that functions in apoptosis and inflammatory responses. Caspase-11 exists as 38 and 43 kDa pro-caspase forms, and as 30 and 10 kDa active forms. Caspase-11 is an initiator caspase that activates other caspases, including Caspase-1 [IL-1β-converting enzyme (ICE)], Caspase-3, and Caspase-7, which are involved in executing apoptosis. When Caspase-11 is deficient, cells are unable to process or secrete Interleukin-1β (IL-1β) and demonstrate defective motility in response to certain stimuli. Unlike other caspases that are expressed constitutively, Caspase-11 expression can be upregulated in cells stimulated with certain activators, e.g., macrophages, lymphocytes and hepatocytes stimulated with lipopolysaccharide.
