Two-color flow cytometric analysis of CRTAM (CD355) expression on activated human peripheral blood mononuclear cells. Human peripheral blood mononuclear cells were stimulated for 4 hours with Phorbol 12-Myristate 13-Acetate (PMA; 10 ng/ml) and Ionomycin (1 µg/ml). The cells were then stained with BD Horizon™ BV421 Mouse Anti-Human CD8 antibody (Cat.No. 562428) and either Alexa Fluor® 647 Mouse IgG2a, κ Isotype Control (Cat. No. 565357; Left Plot) or Alexa Fluor® 647 Mouse Anti-Human CRTAM (CD355) antibody (Cat. No. 566645; Right Plot) at 1 µg/test. Two-color flow-cytometric pseudocolor plots showing the correlated expression of CD8 versus CRTAM (CD355) [or Ig Isotype control staining] were derived from gated events with the forward and side light-scatter characteristics of viable human peripheral blood leucocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

- An isotype control should be used at the same concentration as the antibody of interest.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The CR24.1 monoclonal antibody specifically recognizes Class I MHC-restricted T cell-associated molecule (CRTAM) which is also known as CD355, Class I-restricted T cell-associated molecule, or Cytotoxic and regulatory T-cell molecule. CRTAM (CD355) is a type I transmembrane glycoprotein that belongs to the Nectin family of cell adhesion molecules within the Ig superfamily. It is expressed on activated NK cells, NKT cells, γδ T cells, CD8+ T cells, and by Purkinje neurons of the cerebellum. CRTAM can exist as a homodimer or as a heterodimer when bound to its ligand, Nectin-like molecule 2 (Necl-2), which is encoded by TSLC1 (tumor suppressor gene tumor suppressor in lung cancer-1). Necl-2-CRTAM interactions play important roles in cellular adhesion and migration and can augment IFN-γ production by CD8+ T cells and cytotoxicity by NK cells.
