APC Mouse anti-Human CD23
Flow cytometric analysis of APC-conjugated anti-human CD23 on human periferal blood lymphocytes.  Human peripheral blood lymphocytes were stained with either APC anti-CD23 (clone M-L233, Cat. No. 558690, right panel) or a APC-conjugated mouse IgG1 isotype control (Cat. No. 555751/554681, left panel) and PE anti-CD19 (clone HIB19, Cat. No. 555413).  Flow cytometry was performed on a BD FACSCalibur™ System and the dot plots were derived from the gated events based on light scattering characteristics of lysed whole blood.

Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to APC under optimum conditions, and unconjugated antibody and free APC were removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Product Notices

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
  2. Since applications vary, each investigator should titrate the reagent to obtain optimal results.

M-L233

The M-L233 antibody specifically binds to human CD23, the low affinity receptor for human IgE (FcεRII). CD23 is a type II membrane glycoprotein that is expressed by B cells, monocytes, macrophages, eosinophils, platelets and dendritic cells. CD23 mediates IgE-dependent cytotoxicity and phagocytosis by macrophages and eosinophils. Soluble CD23 (sCD23) can be released by CD23-positive cells as a result of proteolytic cleavage of membrane CD23. Larger fragments of sCD23 (e.g., 25-37 kDa) retain their IgE-binding capacity whereas smaller fragments (ie, ≤ 12 kDa) do not. Soluble CD23 may have immunoregulatory effects on the growth and differentiation of B cells and other cell types.

Allophycocyanin (APC), is part of the BD family of phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 651 nm and an emission maximum (Em Max) at 660 nm. APC is designed to be excited by the Red (627-640 nm) laser and detected using an optical filter centered near 660 nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
