Flow cytometric analysis of CD200R expression on mouse bone marrow cells. BALB/c mouse bone marrow cells were stained with either Alexa Fluor® 647 Rat IgG2a, κ Isotype Control (Cat. No. 557690; dotted line histogram) or Alexa Fluor® 647 Rat Anti-Mouse CD200R (Cat. No. 566345; solid line histogram) at 0.5 µg per test. A histogram showing the expression of CD200R (or Ig Isotype control staining) was derived from gated events with the forward and side light-scatter characteristics of viable myeloid cells. Flow cytometric analysis was performed using a BD™ Canto II Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
  4. The Alexa Fluor®, Pacific Blue™, and Cascade Blue® dye antibody conjugates in this product are sold under license from Molecular Probes, Inc. for research use only, excluding use in combination with microarrays, or as analyte specific reagents. OX-110

The OX-110 monoclonal antibody specifically binds to CD200R which is also known as OX2 receptor 1 (Ox2r) or MOX2 receptor (Mox2r). CD200R is a type I transmembrane glycoprotein that is encoded by Cd200r1 (CD200 receptor 1) and belongs to the C2-set domain containing gene family within the Ig superfamily. CD200R is expressed by hematopoietic cells including monocytes, granulocytes, and T cells.  CD200R functions as an inhibitory receptor when bound by its ligand, CD200. CD200R signaling can inhibit leucocyte functions such as the production of cytokines.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor™ 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 670-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
