Improving and Expanding BD Horizon Brilliant™ ASRs to Enhance Performance

BD has improved the performance and expanded the portfolio for our BD Horizon Brilliant™ Violet analyte specific reagents (ASRs). Beginning with BD Horizon Brilliant™ Violet 605 ASRs, BD has improved lot-to-lot consistency and brightness while offering more conjugate options in that channel. Select BD Horizon Brilliant Violet 421 ASRs are also now offered with this new manufacturing process. BD Horizon BV605 and BV421 ASRs can be used on flow cytometers equipped with a violet laser and appropriate filters, including high color configurations of the BD FACSLyric™ and BD FACSCanto™ systems.*

Clinical Brilliance Remastered

Improved Lot-to-Lot Consistency

BD has significantly improved lot-to-lot consistency and has reduced the allowable manufacturing lot-to-lot variation in brightness by 50% for new BD Horizon Brilliant ASRs. With this improved reagent consistency, you are given greater confidence in the reproducibility of your results.

Illustrative examples from product development are given in Figures 1, 2 and 3.**

Figure 1. Lot-to-lot consistency for CD11c BV605 ASR. There was a 4% variability in median fluorescence intensity (MFI) between development lots 1 and 2. Lysed whole blood gated on monocytes.
Figure 2. Lot-to-lot consistency for CD8 BV605 ASR. There was a 3% variability in MFI between development lots 1 and 2. Lysed whole blood gated on lymphocytes.
Figure 3. Lot-to-lot consistency for CD45 BV605 ASR. There was a 3% variability in MFI between development lots 1 and 2. Lysed whole blood gated on lymphocytes.

Increased Brightness

BD has increased the brightness of the new BD Horizon Brilliant ASRs relative to the original formulation. Additionally, these new BD Horizon Brilliant ASRs are as bright as or brighter than our analogous research use only (RUO) reagents. With improved brightness, dim and rare populations are more easily resolved in multicolor flow panels.

Illustrative examples from product development are given in Figures 4 and 5.**

Figure 4. CD7 BV605 ASR comparison between new (orange) and original (blue) ASR formulation. Based on development lots, the new formulation is on average 70% brighter based upon positive population MFI. Lysed whole blood gated on lymphocytes.
Figure 5. New CD13 BV605 ASR (orange) compared to analogous CD13 BV605 RUO reagent (blue). Based on development lots, the new CD13 BV605 ASR is on average 65% brighter based upon positive population MFI. Lysed whole blood gated on monocytes.

Added Flexibility

With an expanded portfolio of ASRs for violet laser channels, you now have increased flexibility in multicolor panel design. This allows more options to move panel markers to the violet laser channels freeing up the blue and red laser channels.

Illustrative examples from product development are given in Figures 6 and 7.**
