R718 Rat Anti-Mouse CD200R3
R718 Rat Anti-Mouse CD200R3
Multiparameter flow cytometric analysis of CD200R3 expression on mouse bone marrow cells. C57BL/6 mouse bone marrow cells were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142) and stained with PE Rat Anti-Mouse CD49b antibody (Cat. No. 553858/561066) and with either BD Horizon™ Rat IgG2a, κ Isotype Control (Cat. No. 566941; Left Plot) or BD Horizon™ R718 Rat Anti-Mouse CD200R3 antibody (Cat. No. 567842; Right Plot) at 0.5 μg/test. DAPI (4',6-Diamidino-2-Phenylindole, Dihydrochloride) Solution (Cat. No. 564907) was added to cells right before analysis. The bivariate pseudocolor density plot showing the correlated expression of CD200R3 (or Ig Isotype control staining) versus CD49b was derived from gated events with the forward and side light-scatter characteristics of viable (DAPI-negative) cells. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to the dye under optimum conditions and unreacted dye was removed.

Recommended Assay Procedures

BD™ CompBeads can be used as surrogates to assess fluorescence spillover (Compensation). When fluorochrome conjugated antibodies are bound to BD CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cells and BD CompBead to ensure that BD CompBeads are appropriate for your specific cellular application.

Ba13

The Ba13 monoclonal antibody specifically recognizes Cell surface glycoprotein CD200 receptor 3 (CD200R3) which is also known as CD200 cell surface glycoprotein receptor-like b (CD200RLb), CD200 receptor-like 3, and OX-2 Receptor 3. CD200R3 is a single-pass type I transmembrane glycoprotein that is encoded by Cd200r3 (CD200 receptor 3) which belongs to the immunoglobulin superfamily (IgSF). It is expressed on basophils and mast cells that are involved in protective immune responses against parasitic infections and in the development of allergic reactions. Cell surface CD200R3 presents as a disulfide-linked dimeric CD200R-like receptor. The cytoplasmic tail region of CD200R3 contains a positively charged lysine residue which allows this receptor to associate and signal through adaptor proteins such as DAP10, DAP12, CD3z, or FcRγ. CD200R3 functions as an activating receptor to regulate IgE-independent immune responses.

R718
The BD Horizon™ Red 718 (R718) Dye is part of the BD red family of dyes. It is a small organic fluorochrome with an excitation maximum (Ex Max) at 695-nm and an emission maximum (Em Max) at 718-nm. Driven by BD innovation, R718 is designed to be excited by the red laser (627–640-nm) and detected using an optical filter centered near 720-nm (e.g., a 720/40-nm bandpass filter). R718 is a brighter alternative to Alexa Fluor™ 700. R718 is also a bright small molecule alternative to APC-R700 with lower spread into the APC detector. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
