Flow cytometric analysis of Notch3 expression on mouse thymocytes. C57BL/6 Mouse thymocytes were stained with FITC Rat Anti-Mouse CD4 (Cat. No. 553047) and BD Horizon™ BUV395 Rat Anti-Mouse CD8a (Cat. No. 563786) antibodies, and with either PE Rat IgG1, κ Isotype Control (Cat. No. 562292; Dashed Line Histogram) or PE Rat Anti-Mouse Notch3 antibody (Cat. No. 568629; Solid Line Histogram) at 0.25 μg/test. The fluorescence histograms showing Notch3 expression (or Ig Isotype control staining) were derived from gated events with the forward and side light-scatter characteristics of CD4-CD8a- (Left Plot) and CD4+CD8a- (Right Plot) viable thymocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

Recommended Assay Procedures
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The HMN3-133.rMAb is a recombinant monoclonal antibody derived from HMN3-133 hybridoma cells that specifically recognizes an extracellular domain of mouse Neurogenic locus notch homolog protein 3 (Notch3 or Notch 3). Notch3 is a type 1 transmembrane glycoprotein receptor that is encoded by Notch3 (notch 3) which belongs to the Notch family containing four members, Notch1-Notch4. The Notch3 precursor is cleaved in the Golgi and presents as a cell surface heterodimeric receptor. The Notch3 receptor can bind to several membrane-bound ligands including Jagged1, Jagged2 and Delta1. Upon ligand binding, Notch3 undergoes proteolytic cleavage that results in the release of the Notch intracellular domain, NICD. NICD translocates to the nucleus where it can form a transcriptional activator complex with various transcription factors. These multimeric complexes either positively or negatively regulate the expression of multiple genes including those that orchestrate many facets of embryonic development and the subsequent functioning of organ systems such as the hematopoietic, immune, nervous, and cardiovascular systems. In the immune system, Notch3 is reportedly expressed by immature thymocytes, activated peripheral CD4+ and CD8+ T cells, and some dendritic cell subsets, but weakly or not by most types of normal lymphocytes.
