Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C.

Recommended Assay Procedures

We recommend using Cytofix/Cytoperm Cat. No. 554714 to fix and permeabilize cells.

B2C10

The B2C10 monoclonal antibody specifically recognizes Galectin-3 (GAL3/Gal-3) which is also known as, IgE-binding protein (εBP/IgEBP), Carbohydrate-binding protein 35 (CBP35), Galactoside-binding protein (GALBP), or  MAC-2. Galectin-3 is a 30 kDa protein that is encoded by LGALS3 (lectin, galactoside-binding, soluble, 3) and belongs to the galactoside-binding lectins family. It is expressed by a variety of cell types including monocytes/macrophages, dendritic cells, some activated lymphocytes, fibroblasts, epithelial cells, and tumor cells. Galectin-3 has diverse functions and is involved in the induction and regulation of immune responses, inflammation, cell adhesion, chemotaxis, apoptosis, angiogenesis, and fibrosis. Galectin-3 can be differentially expressed in the nucleus, cytoplasm, and surface of cells and can be secreted. Galectin-3 levels can be markedly elevated in the nuclei of proliferating cells suggesting that it may play a role in cell growth regulation. Nuclear galectin-3 has been associated with gene expression and pre-mRNA splicing. Cell surface Galectin-3 reportedly serves as a non-integrin, laminin-binding protein, suggesting its role in cell adhesion to basement membranes. The B2C10 antibody is suitable for immunohistochemical staining of paraffin-embedded tissue sections. The B2C10 antibody reportedly crossreacts with mouse Galectin-3.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
