Two-color flow cytometric analysis of IL-27Rα expression on mouse splenocytes. C57BL/6 mouse splenic leucocytes were stained with FITC Hamster Anti-Mouse CD3e antibody (Cat. No. 553062/553061/561827) and either PE Rat IgG2a, κ Isotype Control (Cat. No. 554689; Left Panel) or PE Rat Anti-Mouse IL-27Rα antibody (Cat. No. 564337; Right Panel). Two-color flow cytometric contour plots show the correlated expression patterns of CD3e versus IL-27Rα (or Ig Isotype control staining) for gated events with the forward and side light-scatter characteristics of viable splenic leucocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ Cell Analyzer System.

The 2918 monoclonal antibody specifically binds to the alpha subunit (IL-27 Receptor alpha/IL-27 Rα) of the mouse Interleukin-27 Receptor complex. IL-27 Rα is a type I transmembrane glycoprotein that belongs to the type I cytokine receptor family. It is also known as WSX-1 and T cell cytokine receptor (TCCR). IL-27 Rα combines with gp130 (CD130) to form a high-affinity cell surface receptor that transduces intracellular signals through the JAK-STAT pathway upon IL-27 binding. IL-27 is a heterodimeric cytokine that is composed of the Epstein-Barr virus-induced gene 3 (EBI3) and IL-27p28 (IL-30) protein subunits. This multifunctional cytokine is produced by activated macrophages, dendritic cells, and epithelial cells. It mediates a broad array of pro- and anti-inflammatory responses. The IL-27 Receptor is expressed by a variety of cell types including naïve T cells and B cells. It is more highly expressed by NK cells and subsets of activated T cells.
