Description

Cell or tissue lysates are a useful control for the western blotting application. A source that can be rich in endogenously expressed proteins, this lysate is convenient to use and provided in a denatured format allowing for immediate loading onto an SDS-PAGE gel.

Preparation And Storage

Do not dilute or boil. Store undiluted at -20°C.

Mouse macrophages (RAW 264.7 cells) were grown to 70% confluencey, then stimulated with 10 ng/ml IFNγ and 1 µg/ml LPS for 12 hours. Western blots of mouse macrophages stimulated with IFNγ and LPS are evaluated for the induction of iNOS and Cox-2 using monoclonal antibodies Cat. No. 610328 and 610203 respectively. Unstimulated mouse macrophages (Cat. No. 611479) show no expression of iNOS or Cox-2.

Recommended Assay Procedures
