Multicolor flow cytometric analysis of TCR Vγ6 expression on newborn Mouse thymocytes. Two-day old BALB/c Mouse thymocytes were isolated and then stained with PE Hamster Anti-Mouse CD3e antibody (Cat. No. 553064), BD Horizon™ BV421 Hamster Anti-Mouse γδ T-Cell Receptor (TCR γδ; Cat. No. 562892), and with either Alexa Fluor™ 647 Mouse IgG1, κ Isotype Control (Cat. No. 565571; Left Plot) or Alexa Fluor™ 647 Mouse Anti-Mouse TCR Vγ6 antibody (Cat. No. 570926; Right Plot) at 0.5 µg/test. BD Via-Probe™ Cell Viability 7-AAD Solution (Cat. No. 555815) was added to cells right before analysis. The bivariate pseudocolor density plot showing the correlated expression of TCR Vγ6 (or Ig Isotype control staining) versus TCR γδ was derived from gated events with the forward and side light-scatter characteristics of viable (7-AAD-negative) CD3e-positive lymphocytes. Flow cytometry and data analysis were performed using a BD FACSymphony™ A5 SE Cell Analyzer System and FlowJo™ Software. Data shown on this Technical Data Sheet are not lot specific.

Recommended Assay Procedures
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The 1C10-1F7 monoclonal antibody specifically recognizes the variable gamma 6 region of the γ subunit of the mouse γδ T cell receptor for antigen, TCR Vγ6 (using the Heilig and Tonegawa nomenclature for mouse TCR γ and δ chains). TCR Vγ6 is encoded by the Trgv6 (T cell receptor gamma, variable 6) gene element. TCR Vγ6 is expressed by a subset of TCR γδ+ thymocytes that appear from approximately embryonic day 15 to birth and express Vγ6 paired with Vδ1 γδ TCR. These invariant TCR Vγ6Vδ1+ T cells home to various tissues including the epithelia of the lungs, peritoneal cavity, uterus, testis, tongue as well as the dermis, lamina propria of the colon, and adipose tissue. They are are programmed to secrete IL-17 upon activation. The 1C10-1F7 antibody has been found useful for immunofluorescent staining and flow cytometric analysis of TCR Vγ6+ T cells from C57BL/6 and BALB/c mice. It was also found useful for immunohistochemical analysis and in vivo blocking studies of TCR Vγ6+ γδ T cell function as well. This antibody is suitable for TCR Vγ6+ thymocyte and γδ T cell separations and analyzing TCR Vγ repertoires expressed by thymocytes, peripheral T cells, and T cell hybridomas in developmental and other experimental model systems. TCR Vγ6 is referred to as TCR Vγ4 using the Garman, Doherty, and Raulet nomenclature for mouse TCR γ and δ chains.
