Multicolor flow cytometric analysis of SLAMF9 expression by Mouse splenic plasmacytoid dendritic cells.  C57BL/6 Mouse splenocytes were treated with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899) to lyse erythrocytes, washed, and preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) [Cat. No. 553141/553142]. The cells were then stained with FITC Hamster Anti-Mouse CD3e (Cat. No. 553062), FITC Rat Anti-Mouse CD19 (Cat. No. 553785), BUV395 Rat Anti-Mouse CD45R/B220 (Cat. No. 563793), BD Horizon™ BV421 Rat Anti-Mouse CD317 (BST2) [Cat. No. 566431] antibodies, and with either BD Horizon™ R718 Mouse IgG1, κ Isotype Control (Cat. No. 567470; Left Plot) or BD Horizon™ R718 Mouse Anti-Mouse SLAMF9 antibody (Cat. No. 569819/569894; Right Plot) at 0.5 µg/test. BD Via-Probe™ Cell Viability 7-AAD Solution (Cat. No. 555815/555816) was added to cells right before analysis. The pseudocolor density plot showing the correlated expression of SLAMF9 (or Ig Isotype control staining) versus CD317 (BST2) was derived by progressive gating on B220/CD45R+ Lineage-negative cells (CD3e-CD19-) with the forward and side light-scatter characteristics of viable (7-AAD-negative) splenic leukocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to the dye under optimum conditions and unreacted dye was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Recommended Assay Procedures

BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cells and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The M349 monoclonal antibody specifically recognizes mouse Signaling lymphocyte activation molecule family member 9 that is also known as SLAM family member 9 (SLAMF9) and CD2 antigen family member 10 (Cd2f10, CD2F-10). SLAMF9 is a type I transmembrane glycoprotein that is encoded by Slamf9 which belongs to the SLAM (CD150)/CD2 subset of the Ig superfamily. SLAMF9 is comprised of an N-terminal extracellular domain that contains an IgV-type domain followed by a IgC2-type domain and a transmembrane region. SLAMF9 has a short lysine- and arginine- rich cytoplasmic domain that lacks Immunoreceptor Tyrosine-based Switch Motifs (ITSMs) characteristic of other SLAM/CD2 subset members. SLAMF9 is variably expressed on plasmacytoid dendritic cells, peritoneal B1 cells and F4/80-low macrophages. SLAMF9 may play a role in the induction of pro-inflammatory cytokines and bacterial clearance.

The BD Horizon™ Red 718 (R718) Dye is part of the BD red family of dyes. It is a small organic fluorochrome with an excitation maximum (Ex Max) at 695-nm and an emission maximum (Em Max) at 718-nm. Driven by BD innovation, R718 is designed to be excited by the red laser (627–640-nm) and detected using an optical filter centered near 720-nm (e.g., a 720/40-nm bandpass filter). R718 is a brighter alternative to Alexa Fluor™ 700. R718 is also a bright small molecule alternative to APC-R700 with lower spread into the APC detector. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
