Biotin Mouse Anti-Rat Erythroid Cells

Data of two three-color analysis of erythroid cells in the rat bone marrow is shown. Cells flushed from LEW femoral bone marrow were simultaneously stained with biotin-conjugated mAb HIS49 (Panel A), PE-conjugated anti-rat CD45 mAb OX-1 (Cat. no. 554878, Panel A), and either FITC-conjugated anti-rat CD71 mAb OX-26 (Cat. no. 554890, Panels B-D) or FITC-conjugated anti-rat CD11b mAb WT.5 (Cat. no. 554982, Panels E-G), followed by Streptavidin-APC (Cat. no. 554067, Panel A).  The two-color contour plot of CD45 vs. HIS49 antigen expression is displayed in Panel A.  The remaining panels display histograms of CD71 or CD11b expression on the gated subpopulations:  B, CD71 on CD45+ HIS49- leukocytes (Region 1); C, CD71 on CD45+ HIS49+ maturing erythroid cells (Region 2); D, CD71 on CD45-HIS49+ erythrocytes (Region 3); E, CD11b on CD45+ HIS49-  leukocytes (Region 1); F, CD11b on CD45+ HIS49+ maturing erythroid cells (Region 2);  G, CD11b on CD45- HIS49+ erythrocytes (Region 3). Flow cytometry was performed on a BD FACSCalibur™ flow cytometry system with data analysis by FlowJo software (TreeStar).

Preparation And Storage

Store undiluted at 4°C. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with biotin under optimum conditions, and unreacted biotin was removed.

Recommended Assay Procedures

The HIS49 antibody can cause aggregation of erythrocytes, which results in depletion of erythrocytes in a concentration-dependent manner.

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

Biotin
Biotin is a ubiquitous co-factor (also known as Vitamin B7) that has many properties that make it extremely useful for molecular biology. Biotin has an extremely high affinity for the Avidin family of proteins (Kd = 10-15 M), making it the perfect tool to link two molecules. Biotin labeled antibodies can be combined with any number of Avidin-conjugated probes in order to customize an assay to a particular need. This is especially useful in the case of magnetic cell separation using streptavidin/magnetic bead conjugates, or in the case of flow cytometry using streptavidin/fluorophore conjugates.
