Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with biotin under optimum conditions, and unreacted biotin was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Recommended Assay Procedures

ELISA Detection: This biotinylated G307-2 antibody is useful as a detection antibody for a sandwich ELISA for measuring rat IL-6 protein levels. Biotinylated G307-2 antibody can be paired with the purified C3-4 antibody (Cat. No. 550644) as the capture antibody, with recombinant rat IL-6 (Cat. No. 557008) as the standard. Biotinylated G307-2 antibody should be titrated (4 - 8 µg/ml) to determine optimal concentration for ELISA detection. To obtain linear standard curves, doubling dilutions of rat IL-6 ranging from ~10,000 to 78 pg/ml are recommended for inclusion in each ELISA plate For specific methodology, please visit the protocols section or chapter on ELISA in the Immune Function Handbook, both of which are posted on our web site, Note: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISA reagents are not recommended for assaying serum or plasma samples. For measuring rat IL-6 in serum or plasma our rat IL-6 BD OptEIA™  ELISA Set (Cat. No. 550319 ) is specially formulated and recommended.

Western Blot: The biotinylated G307-2 antibody has been reported by useful for Western blot. Please note that this application is not routinely tested at BD Biosciences Pharmingen.

Biotin is a ubiquitous co-factor (also known as Vitamin B7) that has many properties that make it extremely useful for molecular biology. Biotin has an extremely high affinity for the Avidin family of proteins (Kd = 10-15 M), making it the perfect tool to link two molecules. Biotin labeled antibodies can be combined with any number of Avidin-conjugated probes in order to customize an assay to a particular need. This is especially useful in the case of magnetic cell separation using streptavidin/magnetic bead conjugates, or in the case of flow cytometry using streptavidin/fluorophore conjugates.
