Two-color flow cytometric analysis of Mouse CD28 expression on mouse splenocytes. BALB/c mouse spleen cells were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142). The cells were then stained with APC Hamster Anti-Mouse CD3e antibody (Cat. No. 553066/561826) and either PE Mouse IgG2b, κ Isotype Control (Cat. No.559529; Left Plot) or PE Mouse Anti-CD28 antibody (Cat. No. 566815; Right Plot) at 0.5 μg/test. A two-color flow cytometric dot plot showing the correlated expression of CD3e versus CD28 (or Ig Isotype control staining) was derived from gated events with the forward and side light-scatter characteristics of viable cells. Flow cytometry and data analysis were performed using a BD LSRFortessa™ Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with R-PE under optimum conditions, and unconjugated antibody and free PE were removed.

The E18 monoclonal antibody specifically recognizes CD28 that is encoded by Cd28 (CD28 antigen) which belongs to the immunoglobulin gene superfamily. CD28 is a ~45 kDa type I transmembrane glycoprotein that can be expressed as a disulfide-linked homodimer. The extracellular region of CD28 contains an IgV-like domain which is followed by a transmembrane region and a cytoplasmic domain. CD28 is expressed on thymocytes, most peripheral CD4+ T cells and CD8+ T cells, NK cells, and plasmacytoid dendritic cells. CD28 functions as a costimulatory receptor upon binding ligands that include CD80 (B7-1) and CD86 (B7-2) that are expressed on antigen presenting cells and B cells. CD28-mediated costimulation augments the proliferation and cytokine production of activated T and NK cells, provides a costimulatory signal for CTL induction, and promotes T cell survival. There is considerable evidence that CD28 is a costimulatory receptor involved in many, but not all, T cell-dependent immune responses. The E18 antibody reportedly blocks the binding of B7 antigens to CD28 and can costimulate T cell activation and proliferation. Spleen cells from a CD28-deficient mouse that was immunized with mouse CD28 were used to generate the hybridoma that secretes the E18 antibody.

R-Phycoerythrin (PE), is part of the BD family of Phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 496 nm and 566 nm and an emission maximum (Em Max) at 576 nm. PE is designed to be excited by the Blue (488 nm), Green (532 nm) and Yellow-Green (561 nm) lasers and detected using an optical filter centered near 575 nm (e.g., a 575/26-nm bandpass filter). As PE is excited by multiple lasers, this can result in cross-laser excitation and fluorescence spillover on instruments with various combinations of Blue, Green, and Yellow-Green lasers. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
