Description

Eukaryotic protein trafficking involves packaging of target molecules into membranous vesicles that bud from a donor compartment, travel to a specific destination, fuse, and release their contents into an acceptor compartment. Here, the protein cargo is retained, while protein components of the vesicular transport machinery are recycled. Components of the transport apparatus are essential for continued specific shuttling of proteins throughout the cell. One such protein, Clathrin, coats vesicles involved in endocytosis and those that mediate transport from the trans-Golgi network (TGN) to endosomes. Clathrin forms a non-covalently bound triskelion that can polymerize into a lattice at the membrane or around an entire vesicle. The triskelion contains a 192 kDa heavy chain and a 22-25 kDa light chain in each leg of its trimeric structure. Vesicles that bud from rims of the Golgi cisternae and from the ER contain a non-clathrin based coat. This coat is an oligomeric complex whose subunits are termed COPs (coatomer proteins). COPI is the golgi- and endosome-associated COP complex, while COPII is the ER-associated coat complex. Clathrin and the COPs serve to define vesicle types and have been extensively studied. However, the proper formation and trafficking of vesicles within the cell is affected by a number of proteins. These components operate at multiple levels of the distribution process, whether a vesicle is part of the endocytic/exocytic pathways or a participant in intracellular protein movement.

Preparation And Storage

Store undiluted at -20°C. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography.
