Western blot analysis of SHIP-1 expression on rat spleen lysate. Rat spleen lysate (Cat. No. 611471) was probed with Purified Mouse Anti-SHIP-1 (Cat. No. 611334) at dilutions of 1:250 (lane 1), 1:500 (lane 2), and 1:1000 (lane 3). SHIP-1 was visualized with HRP Goat Anti-Mouse Ig (Cat. No. 554002), and identified as a band of 150 kDa.

Immunofluorescence analysis of SHIP-1 analysis on ES2 cells. ES2 cells (ATCC® CRL1978™) were probed with Purified Mouse Anti-SHIP-1 (Cat. No. 611334) and visualized with FITC Goat Anti-Mouse Ig (Cat. No. 554001).

Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at -20°C.

  2. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

  5. Sodium azide is a reversible inhibitor of oxidative metabolism; therefore, antibody preparations containing this preservative agent must not be used in cell cultures nor injected into animals. Sodium azide may be removed by washing stained cells or plate-bound antibody or dialyzing soluble antibody in sodium azide-free buffer. Since endotoxin may also affect the results of functional studies, we recommend the NA/LE (No Azide/Low Endotoxin) antibody format, if available, for in vitro and in vivo use.

32/SHIP-1

The 32/SHIP-1 monoclonal antibody specifically recognizes the cytosolic lipid phosphatase, SH2 domain-containing inositol phosphatase 1 (SHIP-1), which is also known as Inpp5d.  SHIP-1 is expressed in cells of the hematopoietic lineage. Cells can phosphorylate and activate SHIP-1 in response to a variety of cytokines or triggering through B or T cell antigen receptors. Activated SHIP-1 hydrolyzes phosphatidylinositol-3,4,5-triphosphate to phosphatidylinositol-3,4-bisphosphate. This ultimately blocks the recruitment of PH-domain containing effector proteins, such as Btk and Akt, and can prevent cellular activation.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
