The 6A6 monoclonal antibody specifically binds to mouse CD96, which is also known as T cell-activated increased late expression protein (Tactile, TACT). CD96 is a type I transmembrane glycoprotein that belongs to the Ig gene superfamily. It is variably expressed on NK cells, and T cells at all developmental stages, including thymocytes and mature αβ T cells, γδ T cells, and NKT cells. CD96 interacts with Nectin-1 (CD111) and CD155 (poliovirus receptor). CD96 can reportedly mediate adhesive interactions between NK cells or T cells and CD155+ target cells. CD96 may compete with CD226 (DNAM 1) for CD155 binding and thus limit NK cell cytokine responses.
The antibody was conjugated to BD Horizon™ BV650 which is part of the BD Horizon Brilliant™ Violet family of dyes. This dye is a tandem fluorochrome of BD Horizon BV421 with an Ex Max of 405-nm and an acceptor dye with an Em Max at 650-nm. BD Horizon BV650 can be excited by the violet laser and detected in a filter used to detect APC-like dyes (eg, 660/20-nm filter). Due to the excitation and emission characteristics of the acceptor dye, there will be spillover into the APC and Alexa Fluor® 700 detectors. However, the spillover can be corrected through compensation as with any other dye combination.
