Flow cytometric analysis of CD182 (CXCR2) on mouse myeloid cells. Bone marrow cells fromC57BL/6 mice were stained with either Purified Rat IgG2b, κ Isotype Control (Cat. No. 555846; dashed line histogram) or Purified Rat Anti-Mouse CD182 (CXCR2) [Cat. No. 567900; solid line histogram] at 0.125 µg/test, followed by PE Goat Anti-Mouse Ig (Multiple Adsorption) (Cat. No. 550589). Fluorescent histograms showing CD182 (CXCR2) expression (or Ig Isotype control staining) were derived from gated cells with the forward and side light-scattering characteristics of intact bone marrow leucocytes. Preparation And Storage

Store undiluted at 4°C. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography.

Product Notices

  2. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
  3. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  4. An isotype control should be used at the same concentration as the antibody of interest.
  5. Sodium azide is a reversible inhibitor of oxidative metabolism; therefore, antibody preparations containing this preservative agent must not be used in cell cultures nor injected into animals. Sodium azide may be removed by washing stained cells or plate-bound antibody or dialyzing soluble antibody in sodium azide-free buffer. Since endotoxin may also affect the results of functional studies, we recommend the NA/LE (No Azide/Low Endotoxin) antibody format, if available, for in vitro and in vivo use.

The V48-2310 monoclonal antibody specifically recognizes CD182 which is also known as C-X-C chemokine receptor type 2 (CXCR2). CD182 is a seven-transmembrane protein and member of the G protein-coupled receptor 1 family that is encoded by Cxcr2 [Chemokine (C-X-C motif) receptor 2]. CD182 is highly expressed on neutrophils and binds to CXCL1 (KC/GRO-alpha), CXCL2 (MIP2), CXCL3 (Dcip1), CXCL5 (LIX), and CXCL7 (NAP-2). CD182 plays a role in neutrophil activation and trafficking.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
