Recommended Assay Procedures

Western blot: Please refer to http://www.bdbiosciences.com/pharmingen/protocols/Western_Blotting.shtml.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. Please refer to www.bdbiosciences.com/us/s/resources for technical protocols.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
  4. Source of all serum proteins is from USDA inspected abattoirs located in the United States.

21/Zyxin

Zyxin is an 82kDa zinc-binding metalloprotein found in focal adhesion plaques. It contains a proline-rich N-terminal domain and a C-terminal domain containing three LIM domains. LIM domains are characterized as zinc-binding motifs involved in gene expression protein-protein interactions. Zyxin interacts with several focal adhesion proteins including α-actinin, Vav, and another LIM domain-containing protein, cCRP. Zyxin associates selectively with an SH3 domain of p95 Vav through its proline-rich N-terminal domain. The focal adhesion protein VASP has also been shown to interact with the 83kDa human homologue of zyxin, p83. Other LIM-containing proteins function as transcription factors or as growth and differentiation factors in embryogenesis. Zyxin and its homologues appear to be involved in the transduction of extracellular signals originating at focal adhesion plaques.

Purified
Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
