SLP-76 (SH2 domain-containing Leukocyte Protein) is a tyrosine phosphoprotein that is involved in the TCR mediated intracellular signaling pathway. It may be involved in the signaling pathways of peripheral blood leukocytes, thymic/splenic cells, and in human T, B, and monocytic cell lines. SLP-76 consists of several motifs that signify its importance in protein-protein interactions involved in intracellular signaling pathways, such as the SH2 domain in the C-terminus, the amino-terminus three 17-amino acid repeats with conserved tyrosine and acidic residues (DYE(S/P)P), and a proline rich region. SLP-76 has been shown to associate with Grb2, PLC-γ, SLAP-130, and Vav-all of which are part of the signaling cascade in T cells. ZAP-70 appears to phosphorylate SLP-76, allowing phosphorylated Vav to bind and, together the SLP-76/Vav complex, cooperatively stimulates NF-AT and IL-2 gene activation. Overexpression of SLP-76 has been reported to increase the TCR-mediated transcription of the IL-2 gene. Furthermore, SHP-1 associates with SLP-76 in normal splenic B cells which suggests a role for SLP-76 in BCR signaling.
