Recommended Assay Procedures
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

X40.huIgG1N297A.rMAb is a chimeric recombinant monoclonal antibody that specifically recognizes keyhole limpet hemocyanin (KLH), an antigen not expressed by mammalian cells. This antibody was engineered using variable domain sequences from the original KLH-specific X40 clone with constant region sequences derived from Human IgG1, κ. Asparagine at position 297 is replaced with alanine (N297A) to further reduce Fc receptor interactions of this reagent. The X40.huIgG1N297A.rMAb can thus serve as a Human IgG1, κ Isotype Control for antibodies with the Human IgG1, κ isotype and N297A mutation.
Note: For flow cytometric analysis, Alexa Fluor™ 647 IgG1, κ N297A Human Isotype Control (Cat. No. 569961) is intended for staining nonpermeabilized cells whereas Alexa Fluor™ 647 IgG1, κ N297A Human Isotype Control (Cat. No. 569794) is intended for staining fixed and permeabilized cells.
