The Protease Inhibitor Cocktail is provided as a powder. Before use, add 1 ml of pure ethanol to obtain a 50X Protease Inhibitor Cocktail. The reconstituted 50X protease inhibitor cocktail will have the following ingredients: 800 µg/ml benzamidine HCl, 500 µg/ml phenanthroline, 500 µg/ml aprotinin, 500 µg/ml leupeptin, 500 µg/ml pepstatin A, and 50 mM PMSF.

Researchers should be aware that the reconstituted 50X protease inhibitor cocktail may be subject to slight insolubility. Solubility will be achieved when preparing the 1X solution in final buffer.

Preparation And Storage

Store the Protease Inhibitor Cocktail either as a powder or its reconstituted form at -20°C.

Recommended Assay Procedures

The Protease Inhibitor Cocktail is added to 1x Insect Cell Lysis Buffer (Cat. No. 554778) (10mM Tris pH 7.5, 130 mM NaCl, 1% Triton™ X-100, 10 mM NaF, 10 mM NaPi, 10 mM NaPPi). The Protease Inhibitor Cocktail should be added fresh to a concentration of 1x just prior to each lysis.

Investigators should note that Phenylmethylsulfonylflouride (PMSF) is known to be toxic:

R23/25                        Toxic by inhalation and if swallowed.

R34                                Causes burns

S4                                Keep away from living quarters.

S9                                Keep container in a well-ventilated place.

S26                                In case of contact with eyes, rinse immediately with plenty of water and seek medical advice.

S36/37/39                Wear suitable protective clothing, gloves and eye/face protection.

S45                                In case of accident or if you feel unwell, seek medical advice immediately.

S60                                This material and its container must be disposed of as hazardous waste.
