Recommended Assay Procedures
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cells and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The M349 monoclonal antibody specifically recognizes mouse Signaling lymphocyte activation molecule family member 9 that is also known as SLAM family member 9 (SLAMF9) and CD2 antigen family member 10 (Cd2f10, CD2F-10). SLAMF9 is a type I transmembrane glycoprotein that is encoded by Slamf9 which belongs to the SLAM (CD150)/CD2 subset of the Ig superfamily. SLAMF9 is comprised of an N-terminal extracellular domain that contains an IgV-type domain followed by a IgC2-type domain and a transmembrane region. SLAMF9 has a short lysine- and arginine- rich cytoplasmic domain that lacks Immunoreceptor Tyrosine-based Switch Motifs (ITSMs) characteristic of other SLAM/CD2 subset members. SLAMF9 is variably expressed on plasmacytoid dendritic cells, peritoneal B1 cells and F4/80-low macrophages. SLAMF9 may play a role in the induction of pro-inflammatory cytokines and bacterial clearance.
