Three-color analysis of erythroid cells in the rat bone marrow. Cells flushed from LEW femoral bone marrow were simultaneously stained with PE-conjugated anti-rat CD71 mAb OX-26 (Cat. no. 554891), biotin-conjugated anti-rat CD45 mAb OX-1 (Cat. no. 554876), and purified mAb HIS49 (Cat. No. 550961), followed by Streptavidin-APC (Cat. no. 554067) and FITC-conjugated anti-mouse IgM mAb R6-60.2 (Cat. no. 553408). The two-color contour plot of CD45 vs. CD71 expression is displayed in Panel A. The remaining panels display histograms of HIS49 antigen expression on the gated subpopulations: B, CD45+ CD71+ bright immature erythroid cells; C, CD45- CD71+ maturing erythroid cells; D, CD45- CD71- erythrocytes; E, CD45+ CD71+ hematopoietic progenitors; and F, CD45+ CD71- leukocytes. Flow cytometry was performed on a BD FACSCalibur™ flow cytometry system with data analysis by FlowJo software (TreeStar).

The HIS49 antibody can cause aggregation of erythrocytes, which results in depletion of erythrocytes in a concentration-dependent manner.
Product Notices
- Since applications vary, each investigator should titrate the reagent to obtain optimal results.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The HIS49 antibody reacts with an antigen found on erythrocytes and erythroid progenitors, but not on granulocytes, monocytes, or lymphocytes.
