Multicolor flow cytometric analysis of CD96 (TACTILE) expression on viable Mouse splenic leukocytes. C57BL/6 Mouse splenic leukocytes were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) [Cat. No. 553142]. The cells were then stained with APC Hamster Anti-Mouse CD3e (Cat. No. 553066; Top Plots), BD Horizon™ BUV395 Mouse Anti-Mouse CD161b,c (NK1.1) antibody (Cat. No. 564144; Bottom Plots) and with either PE Mouse IgG2b, κ Isotype Control (Cat. No. 555058; Left Plots) or PE Mouse Anti-Mouse CD96 (TACTILE) antibody (Cat. No. 572237/572238; Right Plots) at 0.5 μg/test. DAPI Solution (Cat. No. 564907) was added to cells right before analysis. Pseudocolor density plots showing the correlated expression of CD96 (TACTILE) [or Ig Isotype control staining] versus CD3e or NK-1.1 were derived from gated events with the forward and side light-scatter characteristics of viable (DAPI-negative) lymphocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo™ Software. Data shown on this Technical Data Sheet are not lot specific.

Recommended Assay Procedures
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The TX111 monoclonal antibody specifically binds to mouse CD96, which is also known as T cell-activated increased late expression protein (TACTILE). CD96 (TACTILE) is a single pass type I transmembrane glycoprotein that is encoded by Cd96 which belongs to the Ig gene superfamily. It is variably expressed on NK cells, and T cells at all developmental stages, including thymocytes and mature αβ T cells, γδ T cells, and NKT cells. CD96 (TACTILE) expressed on effector T cells and NK cells mediates their binding to Nectin-1 (CD111) and to poliovirus receptor CD155 expressed on target cells. This adhesive interaction can promote the proliferative, cytokine-producing and cytotoxic responses of these effector cells. The TX111 monoclonal antibody can inhibit the binding of CD96 to CD155.
