Multicolor flow cytometric analysis of IRF8 expression in human peripheral blood mononuclear cells (PBMCs). PBMCs were stained with FITC Mouse Anti-Human CD19 antibody (Cat. No. 555412/560994; Top Plots) and BD Horizon™ BV421 Mouse Anti-Human CD123 (Cat. No. 562517/565928; Bottom Plots) antibodies. They were then fixed and permeabilized using the BD Pharmingen™ Transcription Factor Buffer Set (Cat. No. 562574/562725) followed by staining with either PE Mouse IgG1, κ Isotype Control (Cat. No. 559320; Left Plots) or PE Mouse Anti-Human IRF8 antibody (Cat. No. 566373; Right Plots) at 0.06 μg/test. Two-color flow cytometric contour plots showing the correlated expression of CD19 or CD123 versus IRF8 (or Ig Isotype control staining) were derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Cell Analyzer System Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

The U31-644 monoclonal antibody specifically recognizes human Interferon regulatory factor 8 (IRF8) which is also known as Interferon consensus sequence-binding protein (ICSBP or H-ICSBP). IRF8 is a ~50 kDa transcription factor that belongs to the interferon (IFN) regulatory factor (IRF) family which consists of 9 members in both humans and mice. IRF8 is variably expressed in hematopoietic cells including monocytes, macrophages, dendritic cells, and B cells. IRF8 has a conserved N-terminal DNA-binding domain (DBD) and a divergent C-terminal region that functions as the regulatory domain which specifies interactions between IRF8 and other IRF family members, transcription factors, or cofactors. IRF8 functions as a transcriptional activator or repressor depending on the formation of heterodimeric DNA binding complexes with these different partners. IRF8 binds to specific DNA motifs including IFN-stimulated response elements (ISRE) and regulates expression of genes including those that encode IFN-alpha (IFN-α), IFN-beta (IFN-β), and IFN-inducible MHC class I genes. IRF8 plays an important in the development of myeloid cells and B cells. It functions in innate and adaptive immunity by regulating the effector functions of monocytes/macrophages, B cells and dendritic cell subsets. IRF8 participates in the induction of Type I IFNs, TNF, IL-6, and IL-12 expression by leucocytes stimulated with ligands for Toll-like receptors and other pattern recognition receptors (PRRs). IRF8 is also involved in cell cycle regulations and apoptosis and may play a role in tumorigenesis.
