The CD80 antibody, clone L307.4, is derived from the fusion of Sp2/0 mouse cells with spleen cells from a C3H/HeN mouse immunized with B7/SL.2 mouse L-cell transfectant.

The CD80 antibody recognizes a 60-kilodalton (kDa) transmembrane glycoprotein that is a member of the immunoglobulin superfamily. The CD80 antigen acts as the ligand for both the CD28 and CTLA-4 glycoproteins, which are expressed on T lymphocytes. The CD80 antigen interacts with the CD28 and CTLA-4 glycoproteins to provide a stimulus for T-lymphocyte activation. The interaction of the CD80 and CD28 antigens mediates heterotypic cell adhesion and also results in augmentation of T-lymphocyte proliferation and cytokine production. The ligation of the CD80 and CD28 antigens also enhances cytotoxic effectors of cytotoxic T lymphocytes (CTL). Monoclonal antibodies directed against the CD28 or CD80 antigens inhibit alloantigen and mitogen-induced proliferative responses.

R-Phycoerythrin (PE), is part of the BD family of Phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 496 nm and 566 nm and an emission maximum (Em Max) at 576 nm. PE is designed to be excited by the Blue (488 nm), Green (532 nm) and Yellow-Green (561 nm) lasers and detected using an optical filter centered near 575 nm (e.g., a 575/26-nm bandpass filter). As PE is excited by multiple lasers, this can result in cross-laser excitation and fluorescence spillover on instruments with various combinations of Blue, Green, and Yellow-Green lasers. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
