Flow cytometric analysis of human ILT5 (CD85a) expression on human peripheral blood leucocytes. Human whole blood was stained with either Alexa Fluor® 647 Mouse IgG2a, κ Isotype Control (Cat. No. 557715; Left Panel) or Alexa Fluor® 647 Mouse Anti-Human ILT5 (CD85a) antibody (Cat. No. 564469; Right Panel). Erythrocytes were then lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). Flow cytometric contour plots showing the correlated expression patterns of ILT5 (CD85a) [or Ig Isotype control staining] versus side scattered-light (SSC) characteristics were generated from gated events with the forward and side light-scatter characteristics of viable leukocytes. Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System.

- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
- Source of all serum proteins is from USDA inspected abattoirs located in the United States.
- An isotype control should be used at the same concentration as the antibody of interest.

The 222821 monoclonal antibody specifically binds to Immunoglobulin-like transcript 5 (ILT5) which is also known as CD85a (CD85 antigen-like family member A), Leukocyte immunoglobulin-like receptor 3 (LIR-3), and Monocyte inhibitory receptor HL9. ILT5 is encoded by LILRB3 and belongs to the ILT/LTR family in the Ig gene superfamily. ILT5 is expressed by a variety of cell types including monocytes, macrophages, granulocytes, NK cells, osteoclast precursors, and a subset of T lymphocytes. This transmembrane receptor contains intracellular immunoreceptor tyrosine based inhibitory motifs (ITIMs) that can inhibit intracellular signaling. ILT5 can bind to MHC Class I molecules and appears to be involved in regulating immune responses such as NK cell-mediated cytotoxicity.
