The BD Biosciences Custom Technology Team (CTT) works with clients to make both qualitative and quantitative multicolor antibody cocktails for flow cytometry–based immunophenotyping and cell analysis.

These products include conjugated antibodies, compensation controls, buffers, and protocol guidelines. Optimized multicolor flow cytometry cocktails can streamline sample preparation, acquisition, and analysis, and improve standardization between experiments.

BD Horizon™ Dri Multicolor Cocktails

Dri - Blood Tubes

BD offers dried products for flow cytometry, including reagents and control cells. Designed for specific experiment requirements, BD Horizon Dri Multicolor Cocktails are pre-aliquoted formats which can reduce variability between experiments, making these reagents ideal choices for multisite or longitudinal studies. A wide variety of reagents and dyes are available for cocktailing, including BD Horizon Brilliant™ dye reagents.* BD Horizon Dri Multicolor Cocktails also have an extended shelf life compared to liquid reagents with stability of ≥1 year when stored at room temperature (4-27ºC). In addition, researchers can easily incorporate these dried down reagents into automated workflow processes, providing greater consistency, precision, standardization and improved lab efficiency.

BD Horizon™ Dri reagents show comparable performance to liquid reagents as demonstrated in Figures 1-4. The results are taken from a multicolor panel used for the identification and characterization of regulatory T cells (Treg) in human PBMCs. The staining observed with the dried reagents (shown in blue, left plot in Figures 1-4) is comparable to the staining observed with liquid reagents (shown in red, center plot in Figures 1-4), and as shown in the overlay plots (right plot in Figures 1-4).

Lyophilized Products

BD offers lyophilized products for flow cytometry, including reagents and control cells. Designed for specific experiment requirements, lyophilized reagents in pre-aliquoted formats can reduce variability between experiments, making these reagents ideal choices for multisite or longitudinal studies. Lyophilized reagents also have an extended shelf life compared to liquid reagents. In addition, researchers can easily incorporate lyophilized reagents into automated workflow processes, providing greater consistency and improved lab efficiency.

Researchers simply add sample—cells, blood, or other liquids—to rehydrate the biomolecules, and immediately begin experimental analysis. Lyophilized products can be packaged in many formats such as tubes, plates, and single vials.

Studies have also demonstrated that lyophilized reagents retain functionality equivalent to liquid reagents. Using standard protocols for antigen-specific activation of peripheral blood lymphocytes, and assessment of response using cytokine flow cytometry, a strong correlation in activity was found between lyophilized antibody cocktails with liquid cocktails.

Lyophilized cells are excellent as staining, instrument setup, or gating controls. In this experiment, freshly fixed, permeabilized, and stained cells were compared to lyophilized cells that were stained with conjugates to CD69 and IFN-γ (left and middle). Freshly prepared cells were also compared to cells that were stained and then lyophilized. There was virtually no difference in signal intensity or cellular events between these three preparations.
