Analysis of CDX-2 on human colorectal adenocarcinoma cell line DLD-1. DLD-1 cells (ATCC CCL-221) were fixed BD Cytofix™ (Cat. No. 554655) and permeabilized with the BD™ Phosflow Perm/Wash Buffer I (Cat. No. 557885), then stained with Alexa Fluor® 647 Mouse anti-CDX-2 (solid line)  or Alexa Fluor® 647 Mouse IgG1 · Isotype Control (clone MOPC-21, Cat. No. 557732) (dashed line). This antibody also works in BD™ Phosflow Perm Buffer I and II. Flow cytometry was performed on a BD LSR™ II flow cytometry system.

Analysis of CDX-2 on spontaneously differentiated embryonic stem cells.  Human embryonic stem cells (H9; WiCell, Madison, WI) were spontaneously differentiated in a 6-well plate for 3 weeks.  Cells were fixed with BD Cytofix™ (Cat. No. 554655) and permeabilized with the BD™ Phosflow Perm/Wash Buffer I (Cat. No. 557885), then stained with Alexa Fluor® 647 Mouse anti-CDX-2 and PE Mouse anti-Oct3/4 (clone 40/Oct-3, Cat. No. 560186).

Preparation And Storage

The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

M39-711

The M39-711 monoclonal antibody reacts with caudal-type homeobox transcription factor 2 (CDX-2).  In embryonic development, CDX-2 is first expressed in the outer layer of the morula and later in the trophoblast, but not in embryonic stem cells.  Along with Oct3/4, it is involved in the segregation of the trophoectoderm lineage from the inner cell mass.  In the adult, CDX-2 expression is limited to the mucosa of the small and large intestines, where it regulates the synthesis of several intestine-specific proteins.  Loss of CDX-2 expression is observed in some colorectal carcinomas, while gain of CDX-2 expression has been observed in gastric tumors and acute myeloid leukemias.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
