T cells play a key role in the adaptive immune response to a variety of pathogens. However, this response must be carefully regulated to prevent an excessive response. T cells are central to the study of infectious and autoimmune diseases. T cells can be identified by extracellular phenotyping markers and cytokine profiles.

Technologies
Multicolor flow cytometry enables identification of T cell subsets and their activity by allowing simultaneous staining of surface markers and intracellular cytokines or signaling molecules. Complementary technologies for determination of quantity or frequency of cytokine-producing cells include BD Cytometric Array (CBA), ELISA, ELISPOT, and in vivo capture assays.
