Each dot on the plot represents an S. equi cell counted by the BD FACSMicroCount. The result was 3.92 x 109 counts/mL on the BD FACSMicroCount with a corresponding plate count result of 3.40 x 109 cfu/mL.

Here the BD FACSMicroCount results directly correlate with results from traditional methods (eg, plate count, color changing unit, hemacytometer). Time to result was less than 5 minutes per sample.

Whole human blood was stained in a four-color assay for the CD4+ regulatory T cell (Treg) phenotype using the cocktail from the BD Human Regulatory T Cell Sorting Kit (CD4 PerCP-Cy™5.5, CD25 PE, and CD127 Alexa Fluor® 647) (Cat. No. 560753). CD45RA was not included. Anti-CD3 FITC (clone UCHT1) was added to gate the T cells. Samples were acquired on a BD Accuri® flow cytometer. Data was transformed using prior derived FSC-A and SSC-A parameters, then compensated against single-color controls using FlowJo™ Mac® version 9.1. The CD3+CD4+ T cells were gated in Boolean fashion, then interrogated for CD25+CD127low phenotype.
