ABC Transporter Vesicle Assay: Comparison of Cold (LC-MS/MS) and Hot (Radiolabeled) Methods

Abstract
The increasing clinical evidence of transporter-mediated drug-drug interactions has revealed the important role of drug transporters in determining pharmacokinetics, drug efficacy and safety profiles. The guidelines recently published by International Transport Consortium (ITC) and European Medicines Agency (EMA) outline the requirements and suitable in vitro systems for studying drug interactions with these transporters. A variety of in vitro models has been widely applied to access and predict transporter-involved drug elimination and drug-drug interactions in vivo. Inside-out vesicles prepared from insect cells infected with baculovirus expressing ABC efflux transporters have been widely applied to directly evaluate ABC transporter substrate and inhibitor specificity. Due to the lack of robust analytical detection methods, the application of inside-out vesicles in characterizing drug interaction with ABC transporters has been largely limited to radiolabeled chemicals or inhibitory screening assays.
This webinar will focus on the development of an LC-MS/MS detection method using liquid chromatography-tandem mass spectrometry for characterizing drug interactions with two major ABC transporters using the vesicle uptake assay, e.g. human MRP2 and human BCRP. The method was validated by comparing the selected substrate kinetics and inhibitor IC50 generated using both the cold and hot methods.

Abstract
BD™ Cytometric Bead Array (CBA) kits and flex sets are ideal for measuring a variety of soluble and intracellular proteins, including cytokines, chemokines, and growth factors involved in the immune response. By using a broad dynamic range of fluorescence detection and antibody-coated beads, up to 30 proteins can be quantified from a single tube. These assays have typically been performed in a flow cytometry core lab or on a departmental flow cytometer with a dedicated operator. Now the BD Accuri™ C6 personal flow cytometer brings this powerful application to the laboratory benchtop.
Join BD scientist Maria Dinkelmann, PhD, as she discusses the use of BD CBA on the BD Accuri C6. Dr. Dinkelmann will survey the kinds of assays and experiments that can be run, using data examples from the BD CBA Th1/Th2/Th17 kit as well as a panel of immunology flex sets. She will also describe the workflow for performing the assay, data collection, and analysis using FCAP Array™ v3.0.1 software.
