New Brilliant Reagents

A new BD Horizon Brilliant™ Ultraviolet formulation is now available

  • Similar excitation and emission spectral properties with less residual spillover into the BUV395 detector.
  • Better long-term performance stability with comparable or better resolution.
  • The existing catalog antibodies will be conjugated to the new BUV-tandem dye formulation and will be given a new catalog number.
  • The BD Horizon Brilliant Ultraviolet (BUV) tandem dyes are now also available in the BD OptiBuild™ portfolio, unlocking hundreds of new specificities.

BD is committed to developing a reagent portfolio that allows you to fully optimize your multicolor flow experiments. To this extent the BD Horizon Brilliant™ Ultraviolet (BUV) tandem dyes have been reformulated. The spectra of the new and original BUV tandem dyes are very similar so the existing laser line and filter sets are still optimal for excitation and detection. The new reagents exhibit similar or brighter signals compared to the previous reagents. There may be some spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced.

To provide consistency across the BD Horizon BUV portfolio, the catalog items with the predecessor dyes will be discontinued in 2019 and replaced with new catalog numbers with the new formulation of the BUV tandem dyes. There will be a period of time when both the new and predecessor versions of the BUV tandem dyes will be available to give you time to transition.

To distinguish the new from original reagents, new formulated BUV reagents will be packaged in a black vial (the original reagents come in amber vials) the catalog numbers for the off the shelf reagents will start with a 612XXX.

The BUV-tandem dyes were reformulated to improve compatibility with the BD OptiBuild™ technology. In addition to the current catalog of off the shelf BUV reagents, hundreds of BUV tandem dye reagents will be available in BD OptiBuild™.

BD Horizon™ BUV496

The new BD Horizon Brilliant™ Ultraviolet 496 (BUV496) is part of the BD Horizon Ultraviolet family of dyes. The spectral properties of the new formulation of BD Horizon BUV496 tandem dye are very similar to the original version. Both tandem fluorochromes have an Ex Max near 350 nm and an Em Max near 496 nm. The original and new BD Horizon BUV496 can be excited by the ultraviolet laser (355 nm) and detected with a 515/30 nm filter with a 450LP. There may be some small spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced. Due to the excitation of the acceptor dye by the violet laser line, there may be spillover into violet laser detectors with filters in the 505–545 nm range. However, the spillover can be corrected through compensation as with any other dye combination.

Comparison of spillover and spread into the BD Horizon BUV395 detector for original BD Horizon BUV496 tandem fluorochromes vs. the new formulation of BD Horizon BUV496 tandem fluorochromes: CD4 conjugates of each fluorochrome were used to stain fresh lysed whole blood and samples were run on a BD LSRFortessa™ X-20 system. The numbers show the rSD of the positive cells in the BD Horizon BUV395 detector.The new BD Horizon BUV496 showed a 33% decrease in the relative spillover (33% vs. 22% absolute spillover) into the BD Horizon BUV395 detector when compared to the original BD Horizon BUV563. There was a negligible change in spillover (less than 1%) into other detectors. We recommend resetting compensation values when using the new version of the dye. In addition, please be aware that absolute spillover values are instrument settings specific and should be determined by each investigator for their instrument.

BD Horizon™ BUV563

The new BD Horizon BUV563 is part of the BD Horizon Ultraviolet family of dyes. The spectral properties of the new formulation of BD Horizon BUV563 tandem dye are very similar to the original version. Both tandem fluorochromes have an Ex-Max near 350 nm and an Em-Max near 563 nm and can be excited by the ultraviolet laser (355 nm). There may be some small spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced. On instruments with a 561-nm yellow-green laser, the recommended bandpass filter is 585/15 nm with a 535-nm long pass to minimize laser light leakage. When the new BD Horizon BUV563 is used with an instrument that does not have a 561-nm laser, a 560/40-nm filter with a 535-nm long pass may be more optimal. Due to the excitation of the acceptor dye by the blue and yellow-green laser line, there may be spillover into blue and yellow-green laser detectors with filters in the 564-620 nm range. However, the spillover can be corrected through compensation as with any other dye combination.


BUV563 Spectra Full - Large

BD Horizon™ BUV661

The new BD Horizon Brilliant™ Ultraviolet 661 (BUV661) is part of the BD Horizon Ultraviolet family of dyes. The spectral properties of the new formulation of BD Horizon BUV661 tandem dye are very similar to the original version. Both tandem fluorochromes have an Ex Max near 350 nm and an Em Max near 661 nm. The original and new BD Horizon BUV661 can be excited by the ultraviolet laser (355 nm) and detected with a 670/25 filter and a 630 nm LP. There may be some small spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced. Due to the excitation of the acceptor dye by the red laser line, there may be spillover into red laser detectors with filters in the 655-685 nm range. However, the spillover can be corrected through compensation as with any other dye combination.

In addition, please be aware that absolute spillover values are instrument settings specific and should be determined by each investigator for their instrument.

BD Horizon™ BUV737

The new BD Horizon BUV737 is part of the BD Horizon Ultraviolet family of dyes. The spectral properties of the new formulation of BD Horizon BUV737 tandem dye are very similar to the original version. Both tandem fluorochromes have an Ex Max near 350 nm and an Em Max near 737 nm. The original and new BD Horizon BUV737 can be excited by the ultraviolet laser (355 nm) and detected with a 740/35 nm filter. There may be some small spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced. Due to the excitation of the acceptor dye by the red laser line, there may be significant spillover into red laser detectors with filters in the 700-720 nm range. However, the spillover can be corrected through compensation as with any other dye combination.


BUV737 Spectra - Large

BD Horizon™ BUV805

The new BD Horizon BUV805 is part of the BD Horizon Ultraviolet family of dyes. There may be some small spillover differences, so we recommend resetting compensation values when using the new version of the dye. Most notably, the spillover into the BD Horizon BUV395 channel will be reduced.

Which BD Horizon Brilliant™ Ultraviolet (BUV) dyes are changing?

All BD Horizon BUV tandem dyes are changing to a new formulation. These include BD Horizon™ BUV496, BD Horizon™ BUV563, BD Horizon™ BUV661, BD Horizon™ BUV737, and BD Horizon™ BUV805.

BD Horizon™ BUV395 is not changing.


How will I know if I have the original formulation or the new formulation of the tandem dye?

BUV tandem dyes made with the new formulation have a catalog number starting with 6XXXXX and are packaged in black vials. BUV tandem dyes made with the original formulation have a catalog number starting with 5XXXXX and will remain packaged in amber vials.

BUV395 is not changing but will be transitioned into black vials.

When will the existing catalog of reagents (with original formulation) be discontinued?

The catalog numbers of the original versions of the dyes will be replaced with new catalog numbers containing the new dyes. The catalog numbers of the original formulation of the dye will be discontinued in 2019. To provide you with time to transition to the new version of the dye, there will be a period of time when both the new and original versions of the dye will be available for the off-the-shelf reagents.


I would like to finish up a study using the same reagent I am using now (not the new version of the dye). Can I order large amounts of an existing product prior to the discontinuation?

Yes, please order what you need to finish up experiments. You may also order large batches through BDB_Custom_Orders@bd.com.


Why was the BUV tandem dye reformulated?

The BUV tandem dyes were redeveloped to be compatible with BD OptiBuild™ custom technology. We can now offer more BUV tandem fluorochrome options with the antibodies you need, including more esoteric clone/dye combinations.
