Evaluate
and quantitate endothelial cell invasion using real-time fluorescence
and non-destructive sample preparation.
BD
BioCoat Tumor Invasion Systems provide cells with conditions that
allow assessment of their invasive potential in vitro. Available
as a 24-Multiwell insert plate containing the patented BD FluoroBlok
PET membrane (8.0 µm pore size) uniformly coated with BD Matrigel
Matrix which serves as an authentic reconstituted basement membrane
in vitro. This provides a true barrier to non-invasive cells
while presenting an appropriate protein structure for invading cells
to penetrate the BD Matrigel Matrix and pass through the FluoroBlok
membrane. Only those cells that appear on the underside of the FluoroBlok
membrane are quantitated in real time by fluorescence, providing a signal
that can be directly correlated to cell number. All 24 inserts are part
of a single unit. These proprietary systems are compatible with most
fluorescent plate readers, robots and fluid handlers.

Run Real-Time Tumor Cell Invasion Assays Without
Destroying Your Samples

| HT-1080 or 3T3 cells labeled with the fluorescent
dye DiI were placed in a BD BioCoat Tumor Invasion System in
the presence of chemoattractant (5% Fetal Bovine Serum). Invasion
was measured by detecting the fluorescence of cells invading through
the BD Matrigel Matrix and the pores of the FluoroBlok membrane with
an Applied Biosystems CytoFluor® 4000 plate reader at 530 nm excitation
and 590 nm emission (bottom-reading mode). |

Assay-to-Assay Reproducibility
|
|
|
| A single lot of BD BioCoat Tumor Invasion
Systems was assayed four times at three and four day intervals. The
data is normalized to a migration control to minimize variability
due to cell preparation and/or fluorescent dye uptake. The inter-assay
means for the invasive HT-1080 cells was 4837±602 FU (CV = 12.5%). |
