BD's assay systems utilize automation-compatible multiwell systems and are provided with detailed protocols to decrease time in assay development and increase productivity in drug discovery. Parallel artificial membrane permeability assay (PAMPA) is an in vitro model for drug permeability and passive transport. Caco-2 assays, an intestinal epithelial model, are used to assess in vitro bioavailability, permeability and compound transport. For cancer-related studies, the tumor cell invasion and angiogenesis systems are designed for screening compounds related to tumor cell invasion and endothelial cell migration or invasion, respectively.

Caco-2 Transport Assays

The BD BioCoat™ HTS caco-2 assay system is an integrated, automation-friendly cell environment to promote rapid differentiation of caco-2 cells in vitro for robotic screening of prospective pharmaceuticals for oral bioavailability and absorption. Unique fibrillar collagen-coated inserts used with BD’s specialized media accelerate differentiation of caco-2 cells to 3 to 5 days from the traditional 21 day protocol.

Parallel Artificial Membrane Permeability Assay (PAMPA)

Drug compounds are screened for their oral absorption potential early in the drug discovery process in order to eliminate poor performers and to identify candidates that need to be modified. Parallel artificial membrane permeability assays (PAMPA) have become a very useful tool for predicting in vivo drug permeability and are well-suited as a ranking tool for the assessment of compounds with passive transport mechanisms. Use of the PAMPA assay allows for ranking of compounds into a low or high classification, using UV VIS spectroscopy or LC/MS.

The BD Gentest™ pre-coated PAMPA plate system has been validated for use in PAMPA and comes ready to use in your assay. This stable, pre-coated PAMPA plate is highly reproducible and improves assay predictability over traditional PAMPA. Studies of commercially available compounds demonstrate excellent correlation between permeability and human absorption. Highly absorbed compounds (i.e., antipyrine, caffeine, naproxen, ketoprofen), typically under-predicted by traditional PAMPA, demonstrate high permeability (better prediction) with the BD Gentest™ PAMPA plate system.

Tumor Cell Invasion

BD BioCoat Matrigel™ invasion chambers and BD BioCoat tumor invasion systems are pre-coated with BD Matrigel matrix to provide a consistent coating for studying tumor cell invasion. The BD BioCoat Matrigel invasion chambers are made with individual 8 um, clear PET membranes. They are ideal for lower throughput, endpoint assays. The BD BioCoat tumor invasion systems utilize 24- and 96-multiwell inserts with 8 um BD FluoroBlok membranes. The BD FluoroBlok™ 24 and 96-multiwell insert systems offer all the benefits of BD FluoroBlok individual cell culture inserts in an automation-friendly format compatible with most robots and fluid handlers. These systems save time by eliminating cell scraping and manual cell counting; they are best suited for medium and high throughput endpoint or kinetic assays.

Angiogenesis: Endothelial Cell Migration and Invasion

BD BioCoat angiogenesis systems have biological coatings optimized for use in endothelial cell migration and invasion using 3 um BD FluoroBlok fluorescence blocking membranes. The BD BioCoat angiogenesis system: endothelial cell migration and the BD BioCoat angiogenesis system: endothelial cell invasion allow for medium and high throughput endpoint or kinetic assays by eliminating cell scraping and manual cell counting. The automation-friendly format of BD FluoroBlok 24- and 96-multiwell insert systems are compatible with most robots and fluid handlers. These systems are well suited for screening compounds and IC50 determinations.
